Born in Alabama, John Little moved to Buffalo, New York as a teenager to study at the Buffalo Fine Arts Academy from 1924-1927. He then moved to New York to pursue operatic vocal training and opened a successful textile and wallpaper design business.

In 1933 he enrolled in classes at the Art Students League, and studied with George Grosz and later Hans Hofmann. After serving as an aerial photographer during WWII, he returned to New York and moved into Hofmann's 8th Street Studio, where his neighbors were Lee Krasner and Jackson Pollock.

The 1940s and 1950s were a great period of growth and experimentation for Little. He moved to East Hampton in 1951 and in 1957 helped found the Signa Gallery. He exhibited frequently throughout his life, with solo shows at Betty Parsons Gallery in New York, the California Palace of the Legion of Honor in San Francisco, and A. M. Sachs Gallery in New York.
